SDCCAG8, or Serologically Defined Colon Cancer Antigen 8, is a protein encoded by the SDCCAG8 gene in humans. It is also known as CCCAP (Coiled-Coil and C2 Domain-Containing Protein). This protein is widely expressed, with notable concentrations in the kidney, retina, and cells with cilia.SDCCAG8 is a multi-functional protein involved in several cellular processes. One significant function is its role in the centrosome, which is the cell's primary microtubule-organizing center. It is essential for maintaining centrosome integrity and function, and consequently, for proper cell division.In addition, SDCCAG8 plays a crucial role in maintaining cilia, which are tiny hair-like structures on the cell surface involved in sensing environmental signals. SDCCAG8 is required for ciliogenesis and ciliary function, participating in intraflagellar transport and ciliary cargo delivery.
Mutations in the SDCCAG8 gene are associated with several human diseases. These include Bardet-Biedl syndrome, a genetic disorder characterized by obesity, retinal degeneration, polydactyly, and kidney anomalies. Additionally, SDCCAG8 mutations have been linked to nephronophthisis-related ciliopathies (NPHP-RCs), a group of disorders characterized by end-stage kidney disease and often accompanied by extra-renal indicators, including retinal degeneration and intellectual disability.Understanding the function and regulation of SDCCAG8 can provide insights into the biology of cilia, centrosomes, and associated diseases.
