RT1-B Activators primarily function through immunosuppression or anti-inflammatory mechanisms. Some of these activators, such as Cyclosporine, Rapamycin, and FK506, exert their effects by inhibiting key immunological pathways. Cyclosporine and FK506 inhibit calcineurin, thus reducing IL-2 transcription and T-cell activation, which allows for an increase in the functional activity of RT1-B. Rapamycin, on the other hand, inhibits the mTOR pathway, causing a reduction in T-cell proliferation and B-cell differentiation, which indirectly enhances the function of RT1-B by reducing competition for antigen presentation.
Other RT1-B activators, like Aspirin, Indomethacin, Sulindac, Meloxicam, Celecoxib, Rofecoxib, Etoricoxib, and Parecoxib, are non-steroidal anti-inflammatory drugs (NSAIDs) or selective COX-2 inhibitors. These compounds inhibit the COX enzymes, reducing the production of pro-inflammatory prostaglandins and thromboxanes. This decreases inflammation and can indirectly increase the functional activity of RT1-B. Dexamethasone, a glucocorticoid receptor agonist, also falls into this category. By reducing the production of pro-inflammatory cytokines, these agents can indirectly enhance the functional activity of RT1-B by decreasing inflammation and the immune response.
