PKC λ/Δ Inhibitors

PKC λ/ι inhibitors are a diverse group of compounds that primarily target the atypical PKC isoforms λ and ι. These inhibitors are crucial for understanding the biological functions of PKC λ/ι and have possible applications, especially in cancer where these isoforms are often dysregulated. The inhibitors listed above vary in their specificity and potency. Compounds like Aurothiomalate and CID755673 are more selective towards PKC λ/ι, with Aurothiomalate targeting the PB1 domain, crucial for PKC λ/ι's interaction in cellular signaling pathways. CID755673, on the other hand, has been demonstrated to reduce proliferation and induce apoptosis in certain cancer cell lines. Other inhibitors such as Gö6976, ICA-1, and CRT0066101 also exhibit inhibitory effects on PKC λ/ι, albeit with varying degrees of selectivity and potency. ICA-1, for example, is noted for its ability to impair the growth of cancer cells overexpressing PKC ι.
