MASTL Inhibitors

MASTL, an acronym for "Microtubule Associated Serine/Threonine Kinase-Like," is a serine/threonine kinase that plays a pivotal role in the regulation of the cell cycle, specifically during mitosis. The gene encoding MASTL is essential for the proper progression and completion of the cell division process.MASTL's primary role is to phosphorylate and inactivate the phosphatase PP2A-B55 (protein phosphatase 2A), which is responsible for dephosphorylating various proteins during mitosis. By inhibiting PP2A-B55, MASTL ensures that critical proteins remain phosphorylated, which is a requisite for successful mitotic progression, including chromosome condensation, spindle assembly, and cytokinesis. This regulation is crucial for the maintenance of genomic stability, as it stops premature dephosphorylation of proteins that would otherwise lead to errors in chromosome segregation.

The activity of MASTL is tightly controlled, with its kinase activity becoming elevated during mitosis. Dysregulation of MASTL has been associated with chromosomal instability and aneuploidy, conditions often found in cancerous cells. Consequently, aberrant expression or mutations in the MASTL gene have been implicated in the development and progression of cancer. Recent research has also begun to explore MASTL's roles beyond mitosis. It may be involved in the DNA damage response and in the regulation of other cellular processes, although these functions are not yet fully understood.
