Mast Cell Protease 10 Inhibitors

Maskin is a protein that plays a crucial role in the regulation of messenger RNA (mRNA) translation, particularly during early development in eukaryotes. It is a cytoplasmic protein that associates with the translation initiation complex and controls the availability of specific mRNAs for translation, thereby influencing protein synthesis.The name Maskinstems from its function in maskingmRNAs, a process that inhibits their translation at inappropriate times. Maskin achieves this by interacting with eukaryotic translation initiation factor 4E (eIF4E), which is a cap-binding protein essential for the initiation of translation. The cap structure is found at the 5' end of eukaryotic mRNAs and is recognized by eIF4E, marking the mRNA for ribosomal attachment and translation initiation.

Maskin is part of a larger protein complex involving cytoplasmic polyadenylation element-binding protein (CPEB), which binds to specific sequences in the 3' untranslated region (UTR) of mRNAs. In its phosphorylated state, CPEB associates with Maskin, which in turn binds to eIF4E, inhibiting the latter from interacting with eIF4G and thus inhibiting the assembly of the complete translation initiation complex. This effectively silences mRNA translation.The regulation of mRNA translation by Maskin is particularly important in oocytes and early embryonic development. In these contexts, the temporal and spatial expression of proteins is tightly controlled, often through the selective release of mRNAs from translational repression, a process in which Maskin is intimately involved.
