LOXHD1 Activators

LOXHD1, or lipoxygenase homology domains 1, is a protein encoded by the LOXHD1 gene. It is characterized by multiple lipoxygenase homology (LH2) domains, which are typically associated with lipoxygenase enzymes involved in the metabolism of polyunsaturated fatty acids. However, unlike classic lipoxygenases, LOXHD1 does not possess enzymatic activity to catalyze the oxygenation of lipids. Instead, its role is thought to be structural or in molecular binding, given the domain architecture that suggests a potential for protein-protein or protein-lipid interactions.

Expressed predominantly in the hair cells of the inner ear, LOXHD1 is believed to be crucial for the proper function of these sensory cells. The hair cells are responsible for converting mechanical sound vibrations into electrical signals that the brain can interpret as sound. LOXHD1's precise function within the hair cells is not fully understood, but it is thought to be involved in the development, maintenance, or stability of the intricate hair cell stereocilia. These hair-like projections on the surface of hair cells are essential for their mechanosensory function.Mutations in the LOXHD1 gene have been linked to autosomal recessive nonsyndromic hearing impairment. Individuals with mutations in this gene often experience progressive hearing loss, indicating the importance of LOXHD1 for sustained auditory function. The progressive nature of the hearing loss associated with LOXHD1 mutations suggests that the protein may play a role in the long-term preservation of hair cell function or in protection against age- or noise-related cochlear damage.
