Centromere Protein J (CENPJ), also known as Centromere Protein 54 (CENP-54) or SAS-4, is a key protein that plays a critical role in the cell cycle, particularly in centrosome duplication and mitotic spindle assembly. This protein, encoded by the CENPJ gene in humans, is a part of the centromere, a specialized chromosomal region where sister chromatids are connected.CENPJ is essential for the formation of functional cilia, tiny hair-like structures found on the surface of cells. Cilia are necessary for various cellular processes, including signal transduction and locomotion. Thus, mutations in the CENPJ gene can lead to diseases related to ciliary dysfunction, such as Seckel Syndrome and Primary Microcephaly.
Seckel Syndrome, a rare genetic disorder, is characterized by growth delays, intellectual disability, and distinctive facial features. On the other hand, Primary Microcephaly involves a significantly smaller than normal head size, often accompanied by intellectual disability. Both conditions are often caused by mutations in the CENPJ gene, leading to impaired centrosome function and abnormal cell division.CENPJ is also involved in mitosis, the process of cell division. It aids in the formation of the mitotic spindle, the structure that separates chromosomes during cell division. This role is crucial to ensure that each daughter cell receives the correct number of chromosomes.
