INO80B Activators

INO80B Activators are a collection of chemical compounds that indirectly enhance the functional activity of INO80B by influencing the chromatin context in which it operates. Compounds such as Trichostatin A, SAHA (Vorinostat), Scriptaid, RGFP966, and Apicidin function as histone deacetylase inhibitors, leading to an increase in acetylated histones. This hyperacetylation results in a looser chromatin structure, which may facilitate the chromatin remodeling activities of INO80B, such as nucleosome repositioning. Similarly, 5-Azacytidine and Nicotinamide act through inhibition of DNA methylation and sirtuin-mediated deacetylation, respectively, contributing to a chromatin state that could be more amenable to remodeling by INO80B. Anacardic acid, as a HAT inhibitor, modifies histone acetylation patterns, which could also indirectly support INO80B's role in chromatin alteration.

Inthe context of membrane dynamics, Methyl-β-cyclodextrin, by extracting cholesterol, may affect the recruitment of INO80B to chromatin, thus potentially enhancing its activity. Bisphenol A interacts with estrogen receptors and may influence gene expression and chromatin accessibility, indirectly affecting INO80B's function. The selective inhibition of HDACs by MS-275 (Entinostat) and RGFP966 specifically targets histone acetylation, creating a favorable environment for INO80B's action. Furthermore, I-CBP112 disrupts the interaction between acetylated histones and the bromodomain of CREBBP/EP300, which could lead to chromatin states that enhance INO80B-mediated remodeling processes. Collectively, these INO80B Activators, through their targeted effects on chromatin structure and composition, promote conditions that are conducive to the enhancement of INO80B's intrinsic activity in chromatin remodeling without directly interacting with the protein or its immediate substrates.
