FucT-IV, or Fucosyltransferase 4, is an enzyme encoded by the FUT4 gene in humans. It belongs to the family of fucosyltransferases, which are responsible for the catalysis of the transfer of fucose, a hexose sugar, to other molecules, typically proteins and lipids. Fucosylation, the addition of fucose to these molecules, is a type of glycosylation, a post-translational modification that is crucial for various biological processes.FucT-IV specifically catalyzes the addition of fucose to the N-acetylglucosamine residues on glycoproteins and glycolipids, forming Lewis X (Le^x) structures. These structures are important for cell-cell recognition, adhesion, and signaling. The expression of FucT-IV and the resultant fucosylation patterns play significant roles in immune system function, including leukocyte trafficking and inflammation, as well as in the development of the nervous system.
Aberrant expression or function of FucT-IV has been implicated in various diseases. Altered fucosylation patterns can affect immune responses and have been associated with autoimmune diseases, infectious diseases, and cancer. In cancer, changes in fucosylation patterns can influence tumor cell behavior, including proliferation, invasion, and metastasis. These changes can also affect the interaction of tumor cells with the immune system and are of interest for cancer diagnostics. Furthermore, FucT-IV is involved in the synthesis of selectin ligands, which are critical for the interaction of leukocytes with endothelial cells during the immune response. This function highlights its importance in inflammatory processes and immune surveillance.
