FAM9B Activators

FAM9B activators constitute a spectrum of chemical entities that serve to indirectly potentiate the functional activity of FAM9B through various cellular and molecular mechanisms. Epigallocatechin gallate (EGCG) and Forskolin are prime examples; EGCG's inhibitory action on DNA methyltransferases may induce hypomethylation of the FAM9B gene promoter, thereby upregulating FAM9B expression. Concurrently, Forskolin's elevation of intracellular cAMP levels activates PKA, which is capable of phosphorylating substrates that bolster the activity of FAM9B. Another activator, Ionomycin, exerts its effects by increasing intracellular calcium, which in turn activates calmodulin-dependent kinases, potentially culminating in the phosphorylation of proteins that work in concert with FAM9B to enhance its function. Similarly, PMA, through the activation of PKC, can initiate phosphorylation cascades that eventually amplify the activity of FAM9B within its associated signaling pathways.

Furthermore, the activities of FAM9B are modulated by the interaction with various signaling molecules and pathways influenced by chemical compounds like Sphingosine-1-phosphate (S1P), Retinoic acid, and Bisphenol A. S1P, through its receptor-mediated signaling, may foster the phosphorylation of proteins that interact with FAM9B, enhancing its functional activity.
