DR5 activators are compounds that stimulate the activity or enhance the expression of death receptor 5 (DR5). DR5, also known as TNFRSF10B (tumor necrosis factor receptor superfamily member 10B), is a cell surface receptor belonging to the tumor necrosis factor receptor (TNFR) superfamily. This receptor plays a pivotal role in the extrinsic pathway of programmed cell death, also known as apoptosis. The activation of DR5 can lead to the initiation of a cascade of intracellular events culminating in apoptosis, a process critical for maintaining tissue homeostasis, preventing aberrant cell growth, and eliminating harmful cells.

DR5 can be activated by its natural ligand, TNF-related apoptosis-inducing ligand (TRAIL). Upon binding to TRAIL, DR5 undergoes trimerization, which facilitates the recruitment of adaptor proteins and the formation of the death-inducing signaling complex (DISC). This complex activates a series of downstream proteins, including caspases, ultimately leading to the execution phase of apoptosis. DR5 activators, by potentiating its activity or expression, can amplify this pro-apoptotic signaling. Given that the dysregulation of apoptosis is a hallmark of various pathological conditions, understanding the mechanisms of DR5 activation is of paramount importance. The study of DR5 activators offers insights into the intricate molecular events underpinning the extrinsic apoptotic pathway.
