DEHAL1 Inhibitors

DEHAL1 inhibitors consist of a diverse range of chemicals that predominantly target the thyroid hormone synthesis pathway. While several of these inhibitors do not act directly on DEHAL1, their effects on closely related enzymes or processes can significantly impact DEHAL1's function. Key among these are compounds like methimazole, propylthiouracil, and 6-PTU, which operate by inhibiting thyroid peroxidase. This inhibition leads to reduced iodination of tyrosine residues, which in turn results in decreased substrate availability for DEHAL1.

Another class of inhibitors, such as perchlorate, thiocyanate, and potassium thiocyanate, function by hindering iodide uptake into the thyroid gland. As DEHAL1's primary role involves the dehalogenation of iodinated tyrosine residues, any disruption in iodide availability can indirectly impede DEHAL1's activity. This strategy is shared with lithium, which affects multiple stages in thyroid hormone synthesis, thereby influencing DEHAL1 indirectly.
