Calyculin A and Okadaic acid, as inhibitors of protein phosphatases 1 and 2A, prevent the dephosphorylation of proteins, effectively maintaining them in an active phosphorylated state. This strategy of impeding the off-switch for protein activity underscores the complex regulation of cellular processes. LY294002 and H-89 contribute to this regulatory landscape by modulating kinase activity-LY294002 through PI3K inhibition, which can lead to downstream effects on protein activity, and H-89 through inhibiting PKA, thereby potentially inducing compensatory mechanisms that activate other kinases.
