Anaphase promoting complex subunit 16 (APC16) is a component of the anaphase-promoting complex/cyclosome (APC/C), a multi-subunit E3 ubiquitin ligase that plays a pivotal role in cell cycle control by mediating the ubiquitination and subsequent proteasomal degradation of target proteins. The APC/C complex regulates the metaphase-to-anaphase transition and the exit from mitosis into the G1 phase.APC16, being a part of this complex, is integral to its function. It's one of the smallest subunits of the APC/C and is involved in binding other subunits to maintain the stability and functionality of the complex. APC16 is required for the integrity of the APC/C complex and consequently for its ability to mediate the ubiquitination of its target substrates.
The APC/C complex, including APC16, is regulated by several mechanisms such as the binding of coactivator proteins CDC20 and CDH1, phosphorylation, and the spindle assembly checkpoint. This ensures that the APC/C functions at the appropriate time in the cell cycle.Aberrations in the function of the APC/C, potentially involving APC16, can lead to improper cell cycle control and genomic instability, which are hallmarks of many cancers. Furthermore, overexpression of APC/C components has been linked to poor prognosis in several types of cancer.
