Chemical activators of binder of sperm protein homolog 2 can engage various intracellular signaling cascades to promote the protein's activation. Phorbol 12-myristate 13-acetate, a potent activator of protein kinase C (PKC), facilitates the phosphorylation of binder of sperm protein homolog 2, which is a crucial step for its activation. Similarly, forskolin raises levels of cAMP within cells, thereby activating protein kinase A (PKA). Activated PKA is known to phosphorylate various substrates, and in this context, it can target binder of sperm protein homolog 2 for activation. Ionomycin, by increasing intracellular calcium, activates calmodulin-dependent kinases capable of phosphorylating binder of sperm protein homolog 2. Thapsigargin operates through a related mechanism; by inhibiting the sarco/endoplasmic reticulum Ca2+ ATPase (SERCA), it causes a rise in cytosolic calcium that activates kinases which then phosphorylate binder of sperm protein homolog 2.

Lastly, chelerythrine chloride, while typically known as a PKC inhibitor, may induce the activation of alternative kinases that then contribute to the activation of binder of sperm protein homolog 2.
