What formulation would you recommend diluting the antibody in for Westerns and at what dilution?

Thank you for your question. We recommend using 5% nonfat milk + TBST (0.05% Tween-20) for blocking and incubating both primary and secondary antibodies for Western blotting. We recommend a starting dilution of 1:200 (range of 1:100-1:1,000).

Would you recommend 5% BSA or milk powder to dilute the antibody for Western Blot?

Thank you for your question. Our recommended blocking reagent that we also use in our labs is UltraCruz® Blocking Reagent: sc-516214. If you are using dry milk powder or BSA instead, 5 % dry milk powder in TBS-T is generally better for antibodies that bind very prominently and produce a lot of unspecific binding and false positive western blot signals. Antibodies that do not bind as strongly are better incubated with 5 % BSA in TBS-T. Since BSA only contains one type of protein, its blocking effect is not as broad and is better than milk powder if the target protein is not expressed abundantly or the antibody only binds to its target with lower efficiency. It is also important to use only BSA and not milk powder when working with phospho-specific antibodies.

Can this antibody be used for chromosome spreads (PFA)?

Thank you for your question. This antibody has not been validated specifically for chromosome spreads, however it did perform well in IF and IHC with PFA fixed cells and tissue sections. Can TTK (D-8): sc-376842 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, TTK (D-8): sc-376842 has been validated for use in IHC with paraffin-embedded sections.
