I would like to enquire concerning the suitability of sc-13552 for immunoprecipitation. In the brochure we have received together with the purchased antibody there is no indication in the "applications" section that the Ab can be used for IP.

Does the SREBP-2 Antibody (1C6): sc-13552 identify both mature and immature SREBP-2 forms via WB (does it identify 2 different bands?)

Thank you for your question. This antibody, sc-13552 SREBO-2, can recognize both the 125 kDa precursor and the 55-70 kDa COOH-terminal cleaved forms of SREBP-2.

The data sheet says that 1C-6 Ab detects both cleaved and full length forms of srebp2, but only states the one at 125/68kDa, is it possible that it binds to more than one kind of cleaved form of SREBP-2? The 2 extra bands i have are at 50kDa and 65kDa.

Do I have to use mouse-IgGκ BP-HRP as secondary antibody when SREBP-2 antibody(1C6) was used? Is it possible to use mouse-IgG (L+H chain)-HRP as a secondary antibody?

Thank you for your question. You can use any anti-mouse secondary antibody for this SCEBP-2 primary antibody, you do not have to use the mouse-IgGκ BP-HRP.

Rated 3 out of 5 by from Specific but weak signal detected by Western blotThis antibody gives a specific signal corresponding to full-length SREBP2 (120 kDa) and the nuclear transcriptionally active fragment (65 kDa) in total lysates derived from human skin fibroblasts. Unfortunately, the signal is very weak and the appearance of the bands requires high sensitivity ECL and long exposure times. Antibody dilution: 1:400 in TBST + 5% no-fat dry milk, incubation overnight at 4°C.
