In mouse whole liver lysate, the band was observed near the 50~60 kDa is that normal?

Thank you for your question. We have not yet tested this antibody in mouse whole liver lysate. Our Western blot data in human liver lysate was observed around 125 kDa, the molecular weight of the precursor protein. Because this is a mouse monoclonal primary antibody, and you are using it on samples of mouse tissue lysates, we recommend including a secondary antibody control (omit primary antibody). This will confirm whether or not the band you are seeing may be the result of non-specific binding of the anti-mouse IgG secondary antibody alone (or mouse heavy chain IgG).

Does this antibody identifies both mature and immature forms of SREBP1?

Thank you for your question. This mouse monoclonal antibody was raised against amino acids 1119-1147 of SREBP-1 of human origin, but the antibody has not been epitope mapped so further information regarding the epitope location is not available at this time. In Western blot experiments the precursor form is most commonly detected with this antibody.

Thank you for your question. This mouse monoclonal antibody was specific for an epitope mapping between amino acids 1119-1147 at the C-terminus of SREBP-1 of human origin. Protein accession number P36956. Hi !! i am interested to use SREBP-1 (Cat #365513) in liver tissue samples. Does this antibody show 2 bands for precursor and nuclear site detection or i should isolate the nuclear fractions to detect it? Thanks !!

Thank you for your question. We have tested this mouse monoclonal, SREBP-1 Antibody (A-4): sc-365513, by Western blot in human fetal liver tissue extracts and did not observe 2 bands for both the precursor and nuclear p68 protein. This was loading 50 µg of protein for detection. Therefore, we would recommend isolating nuclear fractions to detect the nuclear protein.

Rated 3 out of 5 by from Okay for western blotThis antibody worked pretty good for western blot (rat liver; 25 ug protein; 1:200 dilution; no secondary because the primary is pre-conjugated to HRP). However, the antibody detected a double band at 85 kDa, and the predicted kDa is 125 and 68. So I'm unsure of the reliability, and we won't plan on publishing with this antibody.
