ROCK1 encodes Rho-associated coiled-coil containing protein kinase 1 (Rock-1), a serine/threonine kinase that functions as a principal effector of RhoA to regulate actomyosin contractility and cytoskeletal remodeling. Rock-1 phosphorylates key substrates controlling stress fiber formation, focal adhesion dynamics, and myosin light chain activity, linking Rho/ROCK signaling to cell shape, motility, adhesion, and cytokinesis. Through these processes, ROCK1 contributes to mechanotransduction and tissue architecture, and its dysregulation has been associated with altered invasive behavior and abnormal vascular and fibrotic responses in disease biology. ROCK1 is also implicated in apoptosis and inflammatory signaling contexts, making it a relevant node for dissecting pathway cross-talk in diverse cellular models.
Rock-1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ROCK1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ROCK1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ROCK1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Rock-1 protein expression.
This CRISPR knockout system enables efficient generation of ROCK1-deficient cell models for investigation of Rock-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
