
Nestin Antibody (Rat-401) is a mouse monoclonal IgG1 kappa light chain antibody that detects nestin in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Anti-nestin antibody (Rat-401) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Nestin is a crucial intermediate filament protein predominantly expressed in embryonic central nervous system progenitor cells, playing a vital role in structural integrity and function. Nestin expression is essential during muscle development, where nestin interacts with other intermediate filament proteins such as desmin and vimentin, forming a dynamic network that supports cellular architecture. Nestin′s ability to co-assemble with vimentin or α-internexin into heterodimer coiled-coil structures is significant for the formation of 10 nm intermediate filaments, which are critical for maintaining cellular shape and resilience. Alterations in the intermediate filament consensus rod domain of nestin can disrupt localization in central nervous system precursor cells and radial glial cells, highlighting nestin′s importance in cellular organization and signaling. Nestin serves as a marker for neuroepithelial stem cells, glioma cells, and tumor endothelial cells during periods of rapid growth, indicating nestin′s potential role in cancer biology. During neuron differentiation, nestin localizes to growth cones, suggesting influence on axon elongation and guidance, which is crucial for proper neural network formation. In rat adrenal gland, nestin expression is observed in the zona fasciculata and zona reticularis, as well as in dermatomal cells and myoblasts during early stages of myogenesis, underscoring nestin′s diverse roles in various tissues.
