Should the run the blot in reducing or non-reducing conditions?

Thank you for your question. HSC 70 Antibody (B-6): sc-7298 should recognizes HSC 70 under both reducing and non-reducing conditions, so the choice is up to your experimental design.

Thank you for your question. HSC 70 Antibody (B-6): sc-7298 is specific for an epitope mapping between amino acids 583-601 at the C-terminus of HSC 70 of human origin. It has not been tested for cross-reactivity with other HSP70 family members.

Hi, With this antibody I detect another band at ~120 k, apart from the one at 70k. I wonder if there is possibility of aspecific fixation? I work with a 1:10 000 dilution. Thank you for your help

Thank you for your question. We do not have any information regarding what that 120k band could be as we have not seen it in our in house testing. The antibody should be specific for HSC 70, so I suspect that the band you are seeing is a non-specific band or a result of secondary banding. Rated 5 out of 5 by from Ab with good, clear signal for mouse primary cellsGreat Ab, used it at 1:10000 dilution in 5% skimmed milk in TBS Tween 0.1% (overnight incubation at 4C with agitation) and worked just fine for mouse primary macrophages. Very clear, strong signal, it saturates under a minute of exposure. For reference, I loaded 50 ug of protein per lane, blocked membrane with 5% skimmed milk in TBS-T for 1h, blotted my membrane for another Ab and then reprobed it with this Ab as a loading control the next day. Did 3 10 min washes with TBS-T after both primary and secondary Ab incubation and incubated the secondary Ab for 1h.
