Hint1 Antibody (H-5) is a mouse monoclonal IgG1 antibody that detects Hint1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Hint1, also known as PRKCNH1 or PKCI1, is a 126 amino acid protein that plays a crucial role in cellular processes by localizing to both the cytoplasm and nucleus, where Hint1 exists as a homodimer. Anti-Hint1 antibody (H-5) recognizes a protein containing one HIT (histidine triad) domain primarily involved in hydrolyzing adenosine 5′-monophosphoramidate substrates, including AMP-N-alanine methyl ester, AMP-morpholidate, and AMP-alpha-acetyl lysine methyl ester. Hint1 extends to inhibiting protein kinase C (PKC) activity and influencing apoptosis, which is vital for maintaining cellular homeostasis. Notably, downregulation of p53 and Bax following Hint1 mRNA knockdown indicates a function as a haplo-insufficient tumor suppressor, highlighting significance in cancer biology. Furthermore, promoter hypermethylation of Hint1 has been implicated in hepatocarcinogenesis, suggesting expression alterations may contribute to tumor development.
