FLNC encodes filamin 2, a large actin-binding scaffold that crosslinks F-actin and couples the cytoskeleton to membrane receptors and signaling complexes. Filamin 2 coordinates mechanotransduction, cell shape, and adhesion dynamics by organizing actin networks and interacting with integrins and associated focal adhesion machinery. In striated muscle, FLNC supports sarcomeric and myofibrillar integrity and contributes to force transmission and stress responses. Genetic perturbation of FLNC is associated with cardiomyopathies and myofibrillar myopathies, making it a relevant target for studying cytoskeletal remodeling and muscle disease mechanisms.
Filamin 2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FLNC gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FLNC together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FLNC open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Filamin 2 protein expression.
This CRISPR knockout system enables efficient generation of FLNC-deficient cell models for investigation of Filamin 2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
