CRTAP Antibody (L-24) is a mouse monoclonal IgG2a antibody that detects CRTAP (cartilage associated protein) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). CRTAP is a secreted protein primarily located in the extracellular space, where CRTAP plays a crucial role in collagen post-translational modifications, extracellular fibril assembly, and intracellular trafficking. CRTAP is predominantly expressed in articular chondrocytes and is essential for normal bone formation, as CRTAP contains a signal peptide and a tetratricopeptide-like helical domain. Within the endoplasmic reticulum, CRTAP forms a complex with Gros1 and cyclophilin B, which is vital for the efficient 3-hydroxylation of prolyl residues in Collagen Type I molecules, the major structural proteins of skin and bone. Proper functioning of CRTAP is critical, as mutations in the gene encoding CRTAP can lead to autosomal recessive osteogenesis imperfecta (OI) types 7 and 2B, conditions characterized by bone fragility and a heightened risk of fractures. OI type 7 is a milder form of the disorder, while OI type 2B is associated with neonatal lethality, underscoring the importance of CRTAP in skeletal health and development.
