3–8% Tris-acetate gel로 HAP-1 cell에서 WB을 진행. Ponceau 결과에서 단백질은 잘 transfer되었고, actin 역시 정상적으로 발현이 확인되었습니다. 그러나 target인 BRCA1은 검출되지 않았습니다. 현재 사용 중인 transfer 조건은 400 mA, 90분 (wet transfer)이며, BRCA1이 고분자 단백질이다 보니 transfer 조건을 건드려야할 것 같은데, 조언해주실 부분 있을까요?

Can the antibody be used at 37 oC for 1 h for western blot?

I have tried an aliquote of sc-6954 (Lot-J0622), borrowed from a collegue, and I have got no signal at all in western blot. I wanted this ab because it has lots of citations. Could I get a sample of a different Lot to test it before buying it? Thanks.

What Is the recommended blocking buffer for this antibody(sc-6954)? We have tried LI-COR’s PBS based blocking buffer and 5% BSA in PBST, but have not detected the expected bands, despite detection of phospho-Brca1. We are using several Ewing sarcoma lines

Can BRCA1 Antibody (D-9) use for immunoprecipitation assey in human breast cancer cell lines? We are looking for papers.

Thank you for your question. Yes, this mouse monoclonal, BRCA1 Antibody (D-9): sc-6954 can be used for immunoprecipitation (IP) on human cell lines. We are aware of a couple of publications citing use of the antibody for IP in human breast cancer cell lines. Please refer to PMID's: 27454289 and 25480944.

Rated 1 out of 5 by from Completely Blank BlotSC-6954, Lot# H1122, does not detect BRCA1 on western blot, even with 60µg whole cell extract from human HCT116 cells, 3-8% TA gel, 20% EtOH equilibration, 10' x 25V iblot transfer, and 1:100 O/N incubation. Positive control worked fine.

The wb is completely blank. other antibodies probing for the other protein works well (we do wb everyday). This antibody used to work during my postdoc research. Rated 1 out of 5 by from Failed to stain pachytene spermatocytesNo signal at all at 1:50, 1:100, 1:200 whatsoever. We tried goat and donkey serum - no difference. There were good reviews for the previous Ab C-20. C-20 was rabbite polyclonal and this one is mouse monoclonal, so I guess it skips a lot of signal. D-9 is probably good just for biochemistry. Example is the most recent paper that cited D-9 - in it D9 was used just for pull-downs and WB, but the cytology was done by C-20.
