whether these antibodies were specific to drosophila.

Thank you for your question. We do not recommend this antibody, p53 (DO-1): sc-126, for detection of p53 in drosophila. This antibody is recommended for detection of human p53. We recommend using either our p53 (E-5): sc-74573, or our p53 (C-11): sc-74574, which are specific for drosophila.

I want to use this antibody for IHC on FFPE human brain samples, but I don't find the preferred antigen retrieval method (citrate or EDTA?)

Hi, I noticed that the p53 DO-1 antibody on the abcam website (ab1101) was said 'Does not react with: Mouse, Rat'. But your product datasheet said it can react with p53 from mouse, rat and human origin. Just wondering if this has been tested in house?

Thank you for your question. This mouse monoclonal antibody, p53 (DO-1): sc-126, is a well established clone that is primarily used for detection of p53 of human origin. We are aware of numerous product citations using the DO-1 clone in mouse samples, and we recently obtained some Western blot data in-house in NIH/3T3 whole cell lysate. However, upon closer review this Western blot data will be removed. And we are going to change the species reactivity back to recommended for human only.

I used this antibody for WB to detect endogenous p53 in several cell lines. However, non specific bands were detected around 50-55kDa. My question is what blocking reagents and conditions are suitable to minimize the non specific band  ?

Thank you for your question. We recommend using 5% nonfat milk in TBST (TBS Blotto A: sc-2333) for blocking and antibody incubations. We would recommend running a secondary antibody control, where you incubate your membrane without primary antibody, to see if the non-specific bands are a result of your secondary antibody binding non-specifically. If that is the case, which is fairly common, then as an alternative we would recommend using our Mouse IgG kappa binding protein (m-IgGκ BP-HRP: sc-516102) conjugated to HRP in place of a conventional secondary antibody.
