In western blot I always get an additional band just bellow the main band, I was wondering is this some isoform of p53?

Asked by: Swap
Thank you for your question. We have not seen this band with this antibody, so I have no information on what this could be. Please contact our Technical Service Department or your local distributor for assistance in troubleshooting this antibody.

What is the method used to couple DO-1 to agarose beads and which cross-linker is employed?

Asked by: Frank S
Our antibodies are covalently bound to the agarose beads via primary amines. We use CnBr (Cyanogen Bromide) activated sepharose for the conjugation of our AC products.

Hey! Is it antibody recommended for detection of mice p53 (in the brain)?

Asked by: Meirav
Thank you for your question. We do not currently have in-house data using this antibody to detect p53 in mouse samples, however there are some product citations which reference using this antibody successfully in mouse samples. You can search for these on our website using our Product Citation Search, and limit your search by sc-# and species. https://www.scbt.com/scbt/misc/citationSearch.jsp

Hello! For an endogenous p53 IP, what is the reccomended volume of p53 agarose beads (sc-126 AC) per mg of total protein?

Asked by: CK08
Thank you for your question. You can find instructions on using the agarose conjugate in immunoprecipitation in our recommended protocol under this link: https://www.scbt.com/scbt/resources/protocols/immunoprecipitation-western-blots. Please contact our technical service for further assistance.

For western blotting, which buffer would be the most suitable for this antibody (PBS, TBS, BSA, Milk-based)?

Asked by: Elisa A
Thank you for your question. We recommend using 5% nonfat milk in TBST for blocking, and as a diluent/incubation buffer for both primary and secondary antibodies. Blocking buffer recipe follows in our protocol below. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
