p53 Antibody (DO-1) is an IgG2a kappa mouse monoclonal p53 antibody (also designated tumor suppressor protein p53 antibody or TP53 antibody) that detects the p53 protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. p53 Antibody (DO-1) is available as both the non-conjugated anti-p53 antibody form, as well as multiple conjugated forms of anti-p53 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The 53 protein is a DNA-binding, oligomerization domain- and transcription activation domain-containing tumor suppressor. The p53 protein upregulates growth arrest and apoptosis-related genes in response to stress signals, thereby influencing programmed cell death, cell differentiation, and cell cycle control mechanisms. The p53 Antibody can stain both the nucleus and cytoplasm, as the p53 protein localizes to the nucleus, yet can be chaperoned to the cytoplasm by the negative regulator, MDM2. MDM2 is an E3 ubiquitin ligase that is upregulated in the presence of active p53, where it poly-ubiquitinates p53 for proteasome targeting. p53 fluctuates between latent and active DNA-binding conformations and is differentially activated through posttranslational modifications, including phosphorylation and acetylation. phospho-specific p53 Antibodies can differentiate between phosphorylated and non-phosphorylated p53. Mutations in the DNA-binding domain (DBD) of p53, amino acids 110-286, can compromise energetically-favorable association with cis elements and are implicated in several human cancers. p53 Antibody is therefore an important reagent for cancer research.

Hi I recently purchased this antibody and I am using mouse T22 cells. I don't detect p53 at all. Does it cross-react with mouse ? I tried multiple times now

I tested the antibody in many mouse tissues for WB with secondary antibody recognizing light chain, and I did not get any blot. Besides, it works well in human sample.

Has anyone used this with mouse retinal pigment epithelial cells (RPE)? Have tried a few different IF protocols using this antibody and receive no signal.

Does this Ab (sc-126) contain any glycine, Tris and glycerol? If yes, what are the concentrations of them?

Thank you for your question. Each vial contains 200 µg IgG2a kappa light chain in 1.0 ml of PBS with < 0.1% sodium azide and 0.1% gelatin.

Is P53 DO-1 suitable to detect phosphorylated form  of P53?

How to dilute the sc-126 HRP antibody for WB? And for how long should the WB be incubated?

What's the reactivity of the antibody? It can be used in mouse cell lines by WB?

Thank you for your question. p53 Antibody (DO-1) is recommended for detection of wild type and mutant p53 under denaturing and non-denaturing conditions of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM and can be used in western blotting experiments using murine cell lines.
