TdTIF1 Antibody (G-2) is a mouse monoclonal IgG2b kappa light chain antibody that detects TdTIF1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TdTIF1 (G-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Terminal deoxynucleotidyltransferase (TdT) is a crucial DNA polymerase that catalyzes the addition of deoxyribonucleotides to the 3′-hydroxyl end of DNA primers without requiring a DNA template, which is essential for processes such as DNA repair and replication. The regulation of TdT activity is vital, as specific TdT-interacting factors can positively or negatively influence TdT activity. TdTIF1, also known as DNTTIP1 or TdIF1, is localized in the nucleus and plays a significant role in repressing TdT activity by binding to the Pol β-like region of TdT, effectively masking its DNA-binding site and preventing DNA interaction. TdTIF1 is expressed throughout the body, with the highest levels found in the thymus, and features N-terminal AT-hook motifs that facilitate TdT binding. Beyond its inhibitory role, TdTIF1 also interacts with TReP-132, a transcriptional co-activator of steroidogenic factor 1 (SF-1), enhancing gene expression in steroid-producing cells when bound to SF-1.

Can TdTIF1 (G-2): sc-166296 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Randy McDonald
Thank you for your question. Yes, TdTIF1 (G-2): sc-166296 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
