PLEKHG4 encodes Puratrophin 1, a multidomain protein implicated in neuronal and cytoskeletal regulation through Rho family GTPase signaling. As a putative Rho guanine nucleotide exchange factor–related protein, it is linked to modulation of actin dynamics, membrane trafficking, and neurite architecture, processes central to synaptic maintenance and intracellular transport. PLEKHG4 expression is enriched in the nervous system, supporting roles in neuronal development and circuitry stability. Genetic variation and altered function of Puratrophin 1 have been associated with neurodegenerative and movement-disorder phenotypes, including spinocerebellar ataxia, motivating mechanistic studies in relevant cell models.
Puratrophin 1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PLEKHG4 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PLEKHG4 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PLEKHG4 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Puratrophin 1 protein expression.
This CRISPR knockout system enables efficient generation of PLEKHG4-deficient cell models for investigation of Puratrophin 1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
