Which knock out method is efficient and requires less time? double-nickase knock out? or KO plus HDR knock out? and If i want to select the KO cells , do i need to use the puromycin selection ? or western blot is alone sufficient?

Thank you for your question. Performing Double Nickase and KO +HDR knockout will take approximately the same amount of time for the transfection and selection. You do not have to use the HDR plasmid, but then you will have no selection gene in the KO plasmid alone, so you will have to do single cell selection without narrowing down your cell pool. If I want to knock out CD24 gene in human cell line and finally analysing the CD24 levels using western blot ,which are the products I need to order ?

For the knockout itself you will require the following products: - CD24 CRISPR/Cas9 KO Plasmid (h2): sc-418762-KO-2 - UltraCruz® Transfection Reagent: sc-395739 - Plasmid Transfection Medium: sc-108062 To determine knockout on protein level we recommend using an anti-CD24 antibody such as CD24 Antibody (SN3): sc-19585 as an HRP conjugate or together with m-IgGκ BP-HRP: sc-516102 for secondary detection.
