AEBP1 encodes adipocyte enhancer-binding protein 1, also known as ACLP, an extracellular matrix–associated protein implicated in collagen organization, cell–matrix adhesion, and tissue remodeling. In stromal and fibroblast-like contexts, AEBP1 is linked to pathways that regulate mesenchymal activation, inflammatory signaling, and extracellular matrix turnover, influencing cellular migration and mechanical properties of tissues. Altered AEBP1/ACLP expression has been associated with fibrotic remodeling and tumor microenvironment biology, where it can correlate with changes in stromal composition and immune-related signaling. These features make AEBP1 a useful target for mechanistic studies of matrix-driven phenotypes and cell state transitions.
ACLP/AEBP1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the AEBP1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the AEBP1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the AEBP1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ACLP/AEBP1 protein expression.
This CRISPR knockout system enables efficient generation of AEBP1-deficient cell models for investigation of ACLP/AEBP1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
