CABYR, or calcium-binding tyrosine-(Y)-phosphorylation regulated protein, is a multifunctional protein associated with various cellular processes, particularly within the context of sperm function and fertility. As the name suggests, CABYR has the ability to bind calcium ions, which is essential for its role in calcium signaling pathways that are pivotal during the processes of sperm capacitation and motility.CABYR is predominantly expressed in the male reproductive system, especially within the principal piece of the sperm flagellum. It is known to undergo tyrosine phosphorylation, which is a post-translational modification that can alter the function of the protein, typically as a regulatory mechanism in response to external stimuli. This phosphorylation is a critical step in the cascade of events that lead to the enhanced motility of sperm, known as hyperactivation, and is necessary for successful fertilization.

The protein is thought to interact with components of the fibrous sheath of the sperm tail, which is a unique cytoskeletal structure that is essential for the proper beating of the sperm's flagellum. By binding to this structure, CABYR plays a role in modulating the flexibility and waveform of the sperm tail, thereby affecting its motility patterns.Mutations or alterations in the expression of CABYR have been linked to male infertility, making it a protein of interest in the study of reproductive biology. Its exact mechanisms of action, however, are still under investigation, as researchers strive to fully understand its role in the signaling pathways that govern sperm functionality.
