QUICK LINKS

Electrophoresis sample buffer, typically prepared at a 2X concentration, is a crucial component used in gel electrophoresis techniques, such as SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) and agarose gel electrophoresis. This buffer serves multiple purposes: Denaturation: The sample buffer typically contains denaturing agents like SDS, which disrupts the secondary and tertiary structures of proteins, ensuring that they adopt a linear shape and are uniformly charged based on their mass. Stabilization: Buffers such as Tris-HCl or phosphate are included to maintain a stable pH, ensuring optimal conditions for protein migration. Coloring: Dyes like bromophenol blue or Coomassie blue are often added to the buffer to visualize sample loading during electrophoresis and to monitor the progress of the run. Reducing Agent: In the case of SDS-PAGE, a reducing agent such as beta-mercaptoethanol or dithiothreitol (DTT) may be included to break disulfide bonds and fully denature proteins. The 2X concentration indicates that the buffer is twice as concentrated as what is typically used in the gel. This allows researchers to mix the sample buffer with their sample in a 1:1 ratio, resulting in a final 1X concentration suitable for loading onto the gel. Overall, electrophoresis sample buffer at a 2X concentration is a critical component of gel electrophoresis experiments, ensuring efficient separation and visualization of proteins or nucleic acids based on their size and charge.

沈殿物があり、転倒混和しても溶解しません。また浮遊している粒子がピペットで吸うときに詰まって、正確な量が吸えません。 粒子は沈殿させて上清を吸えば、製品として問題ないのでしょうか?ご教示ください。

Rated 3 out of 5 by from It wasn't necessarily all that usefulI suggest increasing the loading concentration,my sample with 2x loading is too large.

Rated 2 out of 5 by from DisappointedI am unhappy with this product because of 2 reasons: The products gets precipitated and difficult to pipette . The dye concentration is too low making it difficult to track the protein sample.
