

ZNF76 Antibody (A-11) is a mouse monoclonal IgG1 antibody that detects ZNF76 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ZNF76, also known as ZNF523 or Zfp523, is a transcriptional repressor predominantly expressed in the testis, where ZNF76 plays a crucial role in regulating gene expression. ZNF76 is essential for proper transcriptional control of genes encoding small nuclear RNA and selenocysteine tRNA, as ZNF76 is the human homolog of the Xenopus Staf protein, which activates these genes. ZNF76 is localized in the nucleus, where ZNF76 exerts inhibitory function on p53-mediated transactivation, specifically targeting the TFIID complex through interactions with both N and C termini. Post-translational modifications, particularly lysine acetylation and sumoylation, significantly influence ZNF76′s transcriptional repression activity. Acetylation, mediated by p300, leads to a loss of sumoylation and weakened interaction with TFIID, while deacetylation by HDAC1 can restore repressive function. Additionally, splice variants of ZNF76 introduce further complexity to regulatory mechanisms, as these isoforms exhibit differing abilities to interact with TFIID, highlighting ZNF76′s importance in maintaining cellular homeostasis and potential implications in various biological processes.
