This is a membrane-impermeable ethylenediaminofluoresceinyl compound that acts as a high-affinity Zn2+-specific fluorescent probe (Kd = 2.7 nM). It has a low basal fluorescence and the fluorescence intensity increases ~51-fold upon stoichiometric (1:1) binding to Zn2+. it exhibits little affinity towards Ca2+, Na+, Mg2+, or K+. This has been used successfully in monitoring spatio-temporal dynamics of extracellular Zn2+ release in rat hippocampal samples. A cell-permeable, diacetylated form of ZnAF-2 is also available for intracellular use.
