Zinbo-5 is a cell-permeable benzoxazole containing tetradendate fluorescent sensor that serves as a selective and sensitive ratiometric imaging probe for Zn2+ and Cd2+ (by fluorescence I445 nm/I400 nm or by absorbance I376 nm/I337 nm). Zinbo-5 forms a 1:1 complex with zinc (Kd = 2.2 nM, pH 7.2) and displays characteristic changes in both excitation and emission spectra with a ~5-fold increase in fluorescence quantum yield. Zinbo-5 may act to reveal changes in intracellular zinc availability in mouse fibroblasts L(TK-) cells by two-photon excitation fluorescence microscopy and is useful for real time monitoring of Zn2+ in tissues and cells.
