Zic4 Antibody (RY-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects the Zic4 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Zic4 antibody (RY-6) is available as the non-conjugated format. Zic4, or zinc finger protein of the cerebellum 4, is a crucial C2H2 zinc finger transcription factor that plays a significant role in cerebellar development, influencing the formation and maturation of cerebellar granule cells. Zic4 is primarily localized in the nuclei of these cells, where Zic4 regulates gene expression essential for proper cerebellar function. Notably, Zic4 mRNA expression peaks on postnatal day 5 in the developing cerebellum of mice, highlighting Zic4′s importance during early brain development. The Zic family of proteins, including Zic4, is vital for various developmental processes and has been implicated in conditions such as X-linked visceral heterotaxy and holoprosencephaly type 5. Additionally, Zic4 is closely linked to Zic1, another family member located on chromosome 3, which further emphasizes the interconnected roles of these transcription factors in developmental biology.

Can sc-101202: Zic4 (RY-6) monoclonal antibody be used to perform WB in mouse samples? Which dilution do you recommend? (It says 1:100 in one literature https://www.nature.com/articles/s41598-024-71188-7, but it is used for human cancer cells)

Asked by: Joy8
Thank you for your question. sc-101202 is recommended for human samples. There is a 75% homology for mouse. We would not recommend using the antibody for mouse samples.

Can sc-101202: Zic4 (RY-6) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Cweed
Thank you for your question. Yes, sc-101202: Zic4 (RY-6) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
