SEE ALSO...

WT1 Antibody (H-1) is a mouse monoclonal IgM kappa light chain antibody targeting an epitope between amino acids 123-164 within the internal region of the human-origin Wilms′ Tumor 1 protein (WT1), also known as WT33 or EGR-2. Anti-WT1 antibody (H-1) reacts with mouse, rat, and human species and is suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). WT1 is a zinc finger transcription factor that plays a crucial role in kidney and gonadal development by regulating genes involved in cell growth, differentiation, and apoptosis. Predominantly localized in the nucleus, WT1′s cellular location is essential for its function as a transcriptional regulator, and any mislocalization can disrupt gene expression patterns. Mutations or altered expression of WT1 are associated with Wilms′ tumor, a pediatric kidney cancer, as well as other malignancies like leukemia and mesothelioma. Understanding WT1′s nuclear localization and function is important for deciphering the molecular mechanisms of tumorigenesis and normal organ development. WT1 monoclonal antibody (H-1) can effectively detect the expression and intracellular distribution of WT1, providing insights into its role in health and disease. Anti-WT1 antibody (H-1) serves as a valuable tool for advancing research in developmental biology and oncology, potentially aiding in the development of targeted therapies.

I would like to use the WT-1 (H-1) antibody to stain for podocytes in kidney organoids. Do you have any secondary antibody to recommend for fluorescence microscopy?

Asked by: ValeM
Thank you for your question. Here is a link to our secondary antibodies: https://www.scbt.com/scbt/browse/Secondary-Antibodies/_/N-1hoaxk4

Hi, can I ask for the antibody WT1 (H1) insert. Actually I'm looking for WT1 which detect carboxyl Terminus or C-Terminus. Is this is the correct one? to rule out DSRCT (Desmoplastic Small Round Cell Tumours). Can I have the sample for evaluation? TQ

Asked by: RUZA
Thank you for your question. This mouse monoclonal IgM antibody is specific for an epitope mapping between amino acids 123-164 within an internal region of WT1 of human origin. It does not specifically recognize the C-terminus. We do not currently offer any WT1 antibodies with C-terminal epitopes. Please contact our Technical Service Department for more information on how to receive a free 10 ug sample of this monoclonal antibody.

I am using WT1 (H-1): sc-393498 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: cjMara
We recommend performing antigen retrieval with sodium citrate buffer (pH 6.0) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 4 out of 5 by from Works with problemsIt works, but there is always a prominent second band. The second band is very close to the band for WT1 and is thicker and more intense than the WT1 band.

Rated 1 out of 5 by from No IHC or IF staining at all.We acquire a sample and perform IHC and IF on 4um mouse kidney section with M.O.M. kit. None of them works. Podocytes does not have nuclear staining of WT-1, nor has cytoplasmic staining. It`s just completely negative. I really hope the santa Cruz can resume C-19 (sc-192) since it works perfectly on frozen sections with 4% PFA fixation of 20min.
