QUICK LINKS

Western blot stripping buffer is a specialized solution designed for the removal of antibodies and detection reagents bound to membranes in Western blotting experiments. It serves a crucial role in the process by allowing researchers to re-probe membranes with different antibodies, enabling the detection of multiple target proteins from the same sample. The composition of Western blot stripping buffer typically includes chaotropic agents such as urea or guanidine hydrochloride, which disrupt non-covalent interactions between antibodies and their target proteins. Additionally, reducing agents like β-mercaptoethanol or dithiothreitol (DTT) may be included to break disulfide bonds, further facilitating the dissociation of antibody-protein complexes. Some formulations also contain detergents like SDS or Triton X-100 to solubilize proteins and enhance the removal of antibodies from the membrane. The mechanism of action involves the denaturation and solubilization of proteins, leading to the release of antibodies and other bound molecules. After incubation with the stripping buffer, the membrane is typically washed to remove the dissociated proteins and reagents thoroughly. In research, Western blot stripping buffer is invaluable for studying multiple proteins from the same sample or for validating results obtained with different antibodies. By stripping and reprobing membranes, researchers can optimize experimental conditions, confirm the specificity of antibody binding, and obtain comprehensive data from Western blot experiments. Ongoing research focuses on optimizing the composition and efficacy of Western blot stripping buffers to improve their performance and minimize non-specific background signal. Additionally, efforts are underway to develop novel stripping methods that are compatible with sensitive detection techniques and reduce the need for harsh reagents. Overall, Western blot stripping buffer plays a critical role in Western blotting experiments, enabling the efficient reuse of membranes and enhancing the versatility of this widely used protein analysis technique.

I made two times the stripping, but my blots still have the last antibody. Why this could be happening?

I have used your stripping buffer before and it worked fine but this time even using a new batch the PDVF membrane has gone transparent immediately. This is very scary. I tried other stripping buffer and it was fine. Can you please explain this?

During the incubation with the stripping buffer, my membrane becomes transparent. Is this normal?

Asked by: Mathf
Thank you for your question. The change in appearance may be specific to what type of membrane you are using. We have observed some types of membranes become transparent after activation in methanol, so this is possible. Please contact us for Technical Support if you would like to provide more information about what type of membrane you are using,

Can you use this buffer for nitrocellulose or pvdf?

Asked by: woojonglee0917
Thank you very much for the question. Yes, our Western Blot Stripping Buffer is a gentle method for breaking antibody-antigen interactions to allow both, nitrocellulose and PVDF membranes, to be re-probed several times using different antibodies, saving time and conserving samples. THis products is ideal for use with chemiluminescent substrates.

Rated 2 out of 5 by from Mismatching size! Our lab use this stripping buffer for a long time, but there is one thing perplexes us much which is the buffer B (the small one) is too little to pair with buffer A (the big one). We always run out of buffer B before we actually finish the buffer A which means we always need to buy a pair of brand new stripping buffer just for buffer B! It's so annoying though. One suggestion, the company could offer buffer A and B separately for the customers or, why just add up the size of buffer B. The current design of the size is not reasonable, truly.
