Valeryl salicylate, a potent Cox-1 (cyclooxygenase-1) inhibitor, has been found to be an inhibitor of prostaglandin endoperoxide H synthases (PGHS)-1 and -2. The selectivity for Cox-1 versus Cox-2 shows the IC50 values for ovine Cox-1 versus Cox-2 of 0.8 μM versus 15 mM respectively. The inhibition of PGHS-1 took place in a time-dependent manner and was more potent than the inhibition of PGHS-2. Mechanistic studies show that Valeryl salicylate acylates the active site serine of PGHS-1, which prevents arachidonate (sc-200770) binding to the Cox active site and results in suppression of Cox activity.
