Umeclidinium is a potent antagonist of muscarinic acetylcholine receptors, specifically M1-M5 receptors (Kis = 0.05-0.16 nM). It effectively inhibits acetylcholine-induced activation of M3 receptors in CHO cells (IC50 < 10 nM). In human airway smooth muscle cells, Umeclidinium attenuates methacholine-induced calcium release (EC50 = 10 µM) and intranasal administration in mice leads to bronchoconstriction relief (ED50 = 0.02 µg). Umeclidinium bromide, a quaternary ammonium salt, acts as a muscarinic receptor antagonist in the airways, resulting in bronchodilation. By binding to muscarinic receptors, Umeclidinium bromide reduces intracellular calcium levels, leading to relaxation of airway smooth muscle cells.
