    UDP-GlcDH Antibody (A-9) is a mouse monoclonal IgG1 antibody that detects UDP-GlcDH in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). UDP-GlcDH, also known as UDP-glucose 6-dehydrogenase, plays a vital role in converting UDP-glucose to UDP-glucuronic acid, a key precursor in the biosynthesis of glycosaminoglycans, including hyaluronan, heparan sulfate, and chondroitin sulfate. These glycosaminoglycans are essential components of the extracellular matrix, contributing significantly to processes such as signal transduction, cell migration, and cancer progression. UDP-GlcDH functions in the liver, where UDP-glucuronic acid is crucial for the detoxification and excretion of harmful compounds. Structurally, UDP-GlcDH exists as a homohexamer, a configuration highly conserved across various species, demonstrating 97% sequence identity among different mammalian species. Expression of UDP-GlcDH is regulated by factors such as TGFβ, which upregulates expression, and hypoxia, which downregulates expression, highlighting UDP-GlcDH′s potential role in cellular responses to environmental changes.     What blotting conditions were used for the blot? dilution? blocking agent? I used the antibody at 1:1000 in 5% milk/TBST. It was almost clean enough but could be a little better.

    Thank you for your question. You can find dilution recommendations for each antibody on their respective datasheets. Our protocol for western blotting that is used by our labs as well can be found under this link: https://www.scbt.com/resources/protocols/western-immuno-blotting We recommend UltraCruz® Blocking Reagent: sc-516214. Alternatively, a milk-based blocking buffer will produce a cleaner blot while BSA will provide stronger signals. Incubating the blot at a low dilution over night at 4 °C can also help to produce a robust and specific signal in many cases.
