UBE1 is responsible for the first step in the ubiquitin-protein isopeptide bond formation. UBE1 catalyzes the activation of the C-terminal carboxyl group of ubiquitin by forming a high energy thioester bond. This activated ubiquitin is then transferred to the lysine of target proteins via the E2/E3 conjugation cascade. A critical component for the initiation of in vitro conjugation reactions.
