TXNDC4 Antibody (E-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects TXNDC4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TXNDC4 (E-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TXNDC4 protein, also known as ERP44, is a 406 amino acid protein characterized by a thioredoxin domain that plays a crucial role in redox reactions within the cell. Located in the lumen of the endoplasmic reticulum, TXNDC4 is essential for inhibiting the activity of inositol 1,4,5-triphosphate receptor type 1 (IP3R-I) in calcium channels, thereby regulating calcium signaling, which is vital for various cellular processes including muscle contraction, neurotransmitter release, and cell proliferation. Furthermore, TXNDC4 is implicated in oxidative protein folding and the retention of proteins such as Ero1-Lβ and Ero1-Lα within the endoplasmic reticulum, highlighting its importance in maintaining protein homeostasis under stress conditions. Expression of TXNDC4 is upregulated during endoplasmic reticulum stress, indicating a significant role in preserving the intraluminal environment necessary for proper protein maturation and function. Additionally, TXNDC4 features an N-terminal endoplasmic reticulum targeting sequence and a C-terminal endoplasmic reticulum retention signal (RDEL), which are critical for localization and function within the endoplasmic reticulum.

While the staining shown here appears quite dispersed throughout the cytoplasm, I have found my staining of TXNDC4 to be somewhat localised to what looks like the Golgi (as well as general cytoplasm). Has this been seen before? Could it be the dilution?

What is the recommended fixation method for immunofluorescence staining with TXNDC4 (E-6): sc-393687 monoclonal antibody?
