TUG-424 is a potent and selective agonist of the free fatty acid receptor 1 (FFA1) or GPR40. Studies have demonstrated that TUG-424, at a concentration of 100nM, effectively enhances glucose-stimulated insulin secretion in INS-1E cells. FFA1 (GPR40) is predominantly expressed in pancreatic β-cells and is activated by medium and long-chain fatty acids. This receptor plays a role in amplifying glucose-stimulated insulin secretion. CAY10587 is another selective agonist of FFAR1 (GPR40), exhibiting an EC50 value of 32 nM. Notably, it does not affect other related receptors such as FFAR2 (GPR43) or FFAR3 (GPR41). CAY10587, at a concentration of 100 nM, effectively increases glucose-stimulated insulin secretion in INS-1E cells. TUG-424, acting as a potent and selective FFA1 agonist, holds promise in exploring the role of FFA1 in metabolic diseases. It has demonstrated the ability to enhance glucose-stimulated insulin secretion at a concentration of 100 nM in a rat beta-cell line and isolated mouse islets through FFA1 activation.
