Trans-(1S,3R)-ACPD, or trans-1-amino-1,3-dicarboxycyclopentane, is a rigid analog of the neurotransmitter glutamate and a selective agonist of mGluR (metabotropic glutamate receptors). Trans-(1S,3R)-ACPD excites and modulates neuronal activity comparable to the actions of α-kainic acid, producing depolarization of the neuron and subsequent excitation. Seizure and neuronal injury are reported with exposure to Trans-(1S,3R)-ACPD, mediated by mobilization of intracellular calcium stores. Trans-(1S,3R)-ACPD is also described to stimulate phosphoinositide hydrolysis. Trans-(1S,3R)-ACPD is the active isomer of (±)-trans-ACPD preparations. Trans-(1S,3R)-ACPD is an activator of mGluR-2, mGluR-5, mGluR-6.
