QUICK LINKS

TMB (3,3′,5,5′-Tetramethylbenzidine) Substrate Buffer, typically provided as a 1X ready-to-use solution, is extensively employed in biochemical assays, particularly in enzyme-linked immunosorbent assays (ELISAs). The primary role of TMB in these assays is as a chromogenic substrate for the enzyme horseradish peroxidase (HRP), which is often conjugated to antibodies in these tests. The mechanism of action of TMB Substrate Buffer involves the enzymatic conversion of TMB by HRP into a colored product. Initially, TMB is colorless, but upon oxidation by HRP in the presence of hydrogen peroxide (H2O2), it transforms into a blue oxidized form. The intensity of the blue color produced is directly proportional to the amount of enzyme present and, consequently, to the quantity of the target antigen in the assay. This colorimetric change is quantifiable through spectrophotometry, typically at a wavelength of 650 nm for the blue form. For enhanced sensitivity, the reaction can be stopped with an acid, such as sulfuric or hydrochloric acid, which shifts the blue color to yellow, measurable at 450 nm. This shift not only amplifies the signal but also stabilizes it, allowing for more accurate readings. In research, TMB Substrate Buffer is valued for its high sensitivity and specificity. It enables the detection and quantification of various biological molecules, including hormones, proteins, and antibodies, making it indispensable in diagnostics research, quantitative and qualitative analysis of biomarkers, and various other applications where precise measurement of biological interactions is required. The ready-to-use formulation of 1X TMB Substrate Buffer simplifies the assay setup, ensuring consistency and reliability across experiments.
