The Tie-2 Inhibitor 7 can permeate the cell (IC50= 1uM) and inhibit edothelial cell tube formation. Research shows that it can block the Tie-2 ligand, Ang-1 (angiopoietin), which induces Tie2 autophosphorylation and block its downstream signaling with an IC(50)= 0.3 microM. Vascular leakage, suppression of endothelial apoptosis, and vessel regression are strongly inhibited by Ang-1. The Tie 2 receptor plays a crucial role in the normal and pathologic vascular development of epithelial cells.
