TAUT Antibody (A-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects TAUT protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TAUT (A-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TAUT protein, also known as the taurine transporter, is crucial for cellular homeostasis as TAUT facilitates the uptake of taurine, an important organic osmolyte that contributes to antioxidant defense and immune modulation. Located in the plasma membrane, TAUT is a multi-pass membrane protein belonging to the sodium neurotransmitter symporter family, which underscores TAUT′s role in maintaining osmotic balance and cellular integrity. Expression of TAUT is regulated by various factors, including tumor necrosis factor-alpha, which can upregulate TAUT levels, while phosphorylation at Serine 322 serves to downregulate TAUT. Notably, overexpression of TAUT has been shown to confer protection to renal cells against cisplatin-induced nephrotoxicity, highlighting TAUT′s potential therapeutic relevance in kidney injury.

Is it necessary to boil the samples prepared in a loading buffer for 10 min at 70 °C?Or other boiling conditions?

Thank you for your question. Generally, the denaturation condition involves boiling at 95 degrees Celsius for approximately 5 minutes. For multiple transmembrane proteins, special attention needs to be paid to the denaturation conditions of samples when performing WB (Western blot) experiments, as these proteins are prone to aggregation at high temperatures, forming dimers or oligomers that increase their molecular weight, potentially leading to undetected bands or bands appearing at incorrect positions during subsequent detection. Depending on the specific characteristics of the protein, moderate thermal denaturation can be performed within a temperature range of 50°C to 70°C. Some proteins are highly sensitive to heat and have special requirements for the temperature of thermal denaturation; within this temperature range, experimentation can be conducted to find the most suitable denaturation temperature.

Are there any issues with using sc-393036: TAUT (A-11) mouse monoclonal primary antibody on mouse samples?

To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody.
