TACE Antibody (B-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects TACE protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TACE (B-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TACE protein, also known as TNFα converting enzyme, plays a crucial role in the regulation of inflammatory responses by cleaving the membrane-bound precursor of tumor necrosis factor alpha (TNFα) to release its soluble form. This process is vital because TNFα is a key cytokine involved in mediating immune responses and inflammation, and dysregulation can lead to various pathological conditions, including autoimmune diseases and cancer. By facilitating the release of soluble TNFα, TACE not only influences immune cell signaling but also has implications in tumor growth inhibition, making TACE an important target for therapeutic interventions. Anti-TACE antibody (B-6) effectively detects and quantifies TACE in various applications, allowing researchers to explore functional roles and interactions in greater detail, contributing to a deeper understanding of significance in health and disease.

Does this antibody (sc390859) work in tissue obtained from mice? I am interested in brain detection and I have tried other antibodies that work well in rat but not in mouse (although recommended).

Thank you for your question. TACE/ADAM17 Antibody (B-6) is only recommended for detection of TACE precursor of human origin by WB, IP, IF and ELISA. Based on our tests with this antibody it has been found to not be suitable for murine samples.

this mAb can be used for detection by FACS of ADAM17 in jukat?

Thank you for your question. We do have data using this antibody to detect TACE, also designated ADAM17, in Jurkat by Western blot. The conjugated primary antibody is recommended and warrantied for flow cytometry (FCM), however we are still gathering data to show on our website. Check back for updates!

I am interested in your antibody: TACE (B-6): sc-390859. From the description it appears it is meant for TACE precursor, however, I am interested in detecting the active form. Some product reviews appear to indicate it may work. Any further info?

Thank you for the question. TACE (B-6): sc-390859 was raised against amino acids 1-300 mapping at the N-terminus of TACE of human origin. The exact binding epitope has not been mapped, yet. The Pro-peptide comprises aas 18 – 214 and the active Chain 215 – 824. Data seems to indicate the active protein form will be detected. http://www.uniprot.org/uniprot/P78536

Can TACE (B-6): sc-390859 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, TACE (B-6): sc-390859 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

I tested it on protein extracts (30ug) of a cell line of trophoblasts induced to differentiate to an endothelial-like phenotype.
