Suc-Ala-Ala-Ala-pNA is a synthetic peptide used as a chromogenic substrate in biochemistry research for the assay and characterization of proteolytic enzymes, particularly those with trypsin-like activity. When these enzymes cleave the peptide bond after the alanine residue, the p-nitroanilide (pNA) group is released, resulting in a yellow color that can be quantitatively measured using spectrophotometry. This allows for the monitoring of enzyme kinetics and the determination of enzyme activity under various conditions. Additionally, Suc-Ala-Ala-Ala-pNA is employed in the identification and study of protease inhibitors by observing the decrease in colorimetric response upon inhibitor binding. The straightforward and sensitive nature of this assay makes Suc-Ala-Ala-Ala-pNA useful in enzymology for investigating the specificity and regulation of proteolytic enzymes, as well as screening for potential enzyme inhibitors.
